ACE-031: activin receptor research and trial findings

ACE-031 is an engineered soluble activin type IIB receptor fusion protein studied for its effects on muscle. Human trials show why an increase in lean mass is not enough to establish a useful treatment: functional benefit and safety must also be demonstrated.

Identity and formulation

The compound joins an activin receptor component to an immunoglobulin Fc component. It is a larger fusion protein, not a simple short peptide.

Its receptor-trap mechanism interacts with ligands in the activin/myostatin pathway. Calling it only a highly specific myostatin blocker oversimplifies the intervention.

What the studies show

There are directly relevant human data, but the populations and outcomes differ. An early healthy-volunteer study measured body composition after a single exposure. A later Duchenne muscular dystrophy trial was stopped because of safety concerns and did not establish a statistically significant walking benefit.

Study and design Finding What it does not establish
A single ascending-dose study of muscle regulator ACE-031 in healthy volunteers.

Randomized double-blind placebo-controlled single-dose study in 48 healthy postmenopausal women

Reading scope: PubMed abstract and bibliographic record; not full-text review

Investigators observed increases in lean mass and thigh muscle volume, alongside pharmacokinetic and safety measurements. Short-term body-composition changes in this population do not establish improved strength, athletic performance or long-term safety.
Myostatin inhibitor ACE-031 treatment of ambulatory boys with Duchenne muscular dystrophy: Results of a randomized, placebo-controlled clinical trial.

Randomized placebo-controlled study in ambulant boys with Duchenne muscular dystrophy

Reading scope: PubMed abstract and bibliographic record; not full-text review

The study reported trends in body composition and walking measures but was stopped following epistaxis and telangiectasias. The walking trend was not statistically significant; the early stop and vascular adverse-event signals limit efficacy and safety conclusions.
Administration of a soluble activin type IIB receptor promotes skeletal muscle growth independent of fiber type.

Mouse experiments involving soluble activin receptor signaling

Reading scope: PubMed abstract and bibliographic record; not full-text review

The experimental approach increased skeletal muscle mass in mice. Preclinical muscle enlargement does not demonstrate clinical function in humans or validate a commercial preparation.

For the differences between a mechanism, a biomarker and a clinical outcome, see the evidence-reading guide.

How to interpret the findings

Lean mass measured by imaging is an intermediate outcome. A useful muscle treatment should also improve outcomes such as strength, walking, fatigue or daily activities under controlled conditions.

The Duchenne population has a specific disease biology. Neither its preliminary trends nor the healthy-volunteer findings justify extrapolation to bodybuilding, rehabilitation after any injury or routine aging.

Safety context

Nosebleeds and dilated superficial blood vessels were consequential enough to stop the Duchenne study. A statement that no serious adverse event occurred must not be used to hide that reason for stopping.

The short early studies cannot settle delayed effects of interfering with a pathway that has functions beyond skeletal muscle.

Study exposure and units

Exposure schedules in these trials belonged to closely monitored research programs. They are not personal dosing instructions or evidence that repeated use is safe.

Do not compare milligram amounts of a fusion protein with amounts of a short peptide as if potency or biological effects were equivalent.

Match any reported amount or concentration to the exact material, route, population and duration in the original experiment. Read the guide to mass, volume and concentration for the meaning of common units. A calculation cannot establish an appropriate treatment or validate an untested formulation.

Product identity and report checks

A listing should identify the full fusion-protein construct and provide suitable protein-identity and integrity testing. A small-molecule mass or generic peptide purity result is insufficient.

Supplier availability and a certificate cannot resolve the stopped trial, establish functional efficacy or verify that a vial is equivalent to the study material.

Keep the clinical evidence and the supplier evidence separate. Use the report-verification steps to check the issuer and the batch questions to connect a tested sample with the product offered. Compare prices using the delivered-cost method rather than an isolated discount or purity percentage.

Peptide Serum operates a store as well as this research library. The supplier directory contains imported seller information; a listing is not laboratory certification. Read how this library is prepared and evaluate the documentation for the exact lot offered. A seller rating does not establish clinical safety or effectiveness.

Common questions

Did ACE-031 prove a walking benefit in Duchenne muscular dystrophy?

The cited trial reported a trend, not a statistically significant improvement in the walking outcome. The study also stopped early, so a headline of proven functional benefit would overstate it.

Does more lean mass mean stronger muscle?

Not necessarily. Imaging and performance measure different things. The specific endpoint and comparator need to accompany any result.

How to attribute and preserve the context
  1. Place a visible Peptide Serum citation beside the relevant claim, table or passage; do not substitute a general homepage link.
  2. Identify what you reused, and distinguish our compilation or interpretation from an original study or supplier statement.
  3. Keep links and credit to the primary studies, trial registries, supplier listings and other original sources supporting the material.
  4. For price comparisons, preserve currency, units, vial or pack size, sample count, observation dates, exchange-rate date and exclusions.
